Why do I need a 3X normalization minimum?
The xGen™ Pre-Hyb Normalase™ Module chemistry is designed to select a user-defined portion of TruSeq™ adapter libraries for normalization into hybridization capture. Libraries that do not meet the 3X minimum yield threshold (see recommended minimum input ranges in the available protocol) will be under-represented in your pool and will lead to variation in sequencing read distribution. The 3X minimum yield also accounts for the loss of the library that occurs during an average SPRI purification.
