How are IDT HDR donor oligos modified?

We provide 3 options (also see figure here):

  1. Alt-R HDR modification. This is our recommended option for the best HDR performance. It includes a proprietary modification pattern developed through extensive testing to increase donor oligo stability and enable the highest rate of repair.
  2. Phosphorothioate (PS) bond modification. There are 4 PS bonds total; the first 3 and last 3 bases of the HDR donor oligo incorporate these bonds in the sequence. Example: N*N*NNNN…NNNN*N*N (* = phosphorothioate bond; N = DNA base)
  3. No modification. Standard DNA.
